4 Expression Patterns
| Remark | Reporter Gene | Primary Identifier | Pattern | Subcellular Localization |
|---|---|---|---|---|
| Expr11217 | kpc-1 is expressed in numerous neuronal and epithelial cells during dauer. Processes adjacent to the body wall in dauers resemble those of IL2Q 4 dendrites. To determine whether kpc-1 is expressed in the IL2s, we coinjected the Pkpc-1::GFP reporter with a Pklp-6::tdTomato reporter that is expressed exclusively in the IL2s. During nondauer stages, kpc-1 is not expressed in the IL2 neurons (data not shown). In dauers, Pkpc-1::GFP and Pklp-6::tdTomato are coexpressed, indicating that kpc-1 is upregulated in the IL2s during dauer. Nondauer expression was consistently observed in the ventral nerve cord and pharynx with strong expression in the g2 pharyngeal gland cells and vpi pharyngeal intestinal valve cells. | |||
| Other strains-- UL856, UL853 | Expr162 | Strong expression is seen in the excretory cells and canals. Expression in the canals is concentrated in unidentified subcellular structures. Expression is also seen in the terminal bulb of the pharynx, possibly in the g2 gland cell. Expression is most frequently seen in young larvae, but is occasionally seen in embryos and adults. | ||
| Expr11043 | F48A11.1 was expressed specifically in the period immediately preceding each moult. Expression appeared limited to the pharynx and specifically to the glandular cells g1 and g2 (5 nuclei) in the terminal bulb, the three m4 myo-epithelial cells (6 nuclei) in the metacorpus and the three m3 myo-epithelial cells (6 nuclei) in the pro- corpus. No GFP was detected elsewhere in transgenic nematodes despite careful examination of the reproductive tissues and intestine. | |||
| Expr16579 | Using a promoter immediately upstream of the start codon of the eight longest pezo-1 isoforms, we drove expression of GFP in several mesodermal tissues, including the body wall and pharyngeal muscles, pharyngeal glands and valve, and uterine and defecation tissues. Our findings are consistent with those described in a recent preprint by Millet et al. (2021). Based on their unique morphology and location, we identified GFP expression in pharyngeal muscles (pm3-8), pharyngeal glands (dg1, vg1s, vg2s), pharyngeal-intestinal valve (vpi) cells, AMsh, and AMso glial cells. We also tentatively identified pharyngeal neurons M1 and I4. Medially, we saw labeling of the HSN neuron, the vulval muscles, uterine muscles, and intestine. Posteriorly, we saw labeling in cells involved in defection. Based on their morphology and position, we tentatively identified these as rectD, K, F, B, U, and Y (N1⁄421). In addition to GFP-based transcriptional reporters, we used the CRISPR/Cas9-generated AG467 (integrated) strain which used mScarlet to tag the C terminus of PEZO-1 proteins. Imaging of AG467 (N 1⁄4 6) showed PEZO-1 proteins localized to the vicinity of the pharyngeal grinder, to the quadrants of the pharyngeal bulb occupied by the gland cells, and to the tip of the nose of the worm where the amphid sheath is located. We conclude that reporters upstream of long pezo-1 isoforms drive gene expression largely in muscles and other mesodermal tissues, although some neuronal expression was also seen. |
