Reporter gene fusion type not specified. pax-1 expressed in pharyngeal marginal and epithelial cells. (J. Stevenson, A. Chisholm, and S. E. Mango, unpublished data).   Expr4263 pax-1::GFP was activated at around 290 min after the two-cell stage of embryogenesis.
Moreover, neither the dgn-1::GFP promoter reporter nor the rescuing DGN-1::GFP fusion show expression in muscle. Plasmid pJJ516 was made by inserting GFP from pPD114.38 into the HindIII site near the end of the dgn-1 coding sequence. The product contains GFP inserted after residue 575 of DGN-1, with the final seven DGN-1 residues at the C terminus. Expression of DGN-1::GFP is identical to that of the dgn-1::GFP promoter reporter, and DGN-1::GFP rescues the sterility of dgn-1(cg121).   Expr4218 In early (pre-morphological) embryos, dgn-1::GFP expression is evident in many epithelial and neural precursors comprising the outer layer of cells. As elongation begins at comma stage, expression becomes most prominent in several specialized epithelial cells, including pharyngeal e2 and marginal cells, excretory cells, the somatic gonad precursors (SGPs) Z1 and Z4, and rectal epithelial cells. Weaker expression is apparent in hypodermal precursors and neuroblasts along the ventral midline. Pharyngeal expression persists through the L3 larval stage, whereas excretory and rectal cell expression persists throughout development. SGP expression persists in SGP descendants, such as the distal tip cells (DTCs), and increases throughout the gonad during the L4 stage. Variable, generally weak expression is seen throughout larval development in several neurons, although PVP neurons show strong expression throughout development. Transient increased expression occurs in new P cell-derived neurons in the ventral nerve cord in late L1/early L2 stage animals. Variable weak expression is seen in hypodermal cells, principally hyp5 in the head. Preceding the L4/adult molt, expression increases in the vulval epithelium.
Expr4466 Exclusively expressed in the pharyngeal muscle or marginal cells from the embryonic comma stage to adults.
Expr4446 F27E11.3A is expressed in three cells around the nerve ring, possibly neurons, in two cells in the posterior bulb of the pharynx, and in the pharyngeal marginal or muscle cells.
Expr11293 Expressed in pharyngeal marginal cells, some arcade cells, in early larvae: gut.
Expr14868 We observed SMS-5::YFP to be expressed in only two tissues: the spermatheca and the pharynx. In the pharynx, SMS-5::YFP is expressed in the muscle and marginal cells, with an enrichment at cell-cell junctions, and a clear enrichment in the anterior half (corpus) relative to the posterior half (isthmus and terminal bulb). Animals mosaic for the transgene show SMS-5::YFP expression in the marginal cells.
Expr11301 Fairly strong expression all stages (strongest in young larvae), gut, pharyngeal muscle, marginal cells and gland cells, body wall muscle, anal depressor, vulval muscle, spermatheca, about 10 head neurons.
This Expr_pattern is about CeTMIV, an isoform of tmy-1 transcription. To confirm the CeTMIV isoform expression pattern, corresponding tmy-1::gfp vectors were assayed. Similar GFP expressions induced in pharynx and intestines respectively. These results show that the CeTMIV isoform was expressed in the pharyngeal muscles and intestinal cells and establish that the primary promoter region was located within 853 bp upstream of the initial ATG. pretzel stage (author) = fully-elongated embryo (wjc).   Expr1679 The constructs pTMZIV4349 and pTMZIV1957 induced beta-galactosidase expressions in both the pharyngeal muscles and intestinal cells with similar intensities. Specifically, pharyngeal expressions were observed in all the eight muscle cells (m1-m8), one marginal cell (mc), one epithelial cell (e1) and the four cells of the pharyngo-intestinal valve (PIV). The 20 intestinal cells, some of which are binucleated and localized alongside the intestine, posterior of pharynx and anterior of anus were all stained with intense staining occurring at the most posterior end. Intestinal staining was limited only to intestinal cells, although there were slight variations in position of nuclei and staining intensity. Expression was also detected in embryos between gastrulation and the comma stage. At this stage of development, the exact nuclei are difficult to identify, but the positions and topology suggest pharynx and intestines. By the pretzel stage, identification of the different pharyngeal muscles showing expression becomes possible. A similar expression was also observed in the pharynx of males, although the intestinal staining was more restricted to the posterior region. No expression was induced by the further deletion construct pTMZIV1219. In all cases, the most uniform and intense expression patterns were observed between L1 and L2 worms, and were completed by L2. From L3 to adult, there was a reduction in expression intensity. Both pharyngeal and intestinal expressions were evident within six hours after staining.
Expr1808 Only the pharyngeal marginal zone was stained in all larval and adult stages, but not in pre-hatching embryos.
Expr11129 Using transcriptional and translational GFP-based reporters, we identified the pharyngeal marginal cells as the major site of expression of cyp-13A12.
Other strains-- UL257, UL258, UL262, UL263, UL265.   Expr109 Expression begins during embryogenesis and is seen in adults and throughout all larval stages. Strong expression is seen in 3-fold embryos in and around the pharynx. In larvae and adults a number of small nuclei stain at the anterior base of the terminal bulb, and is sometimes accompanied by cytoplasmic staining in the isthmus. This diffuse staining is stronger in larvae and fades in adults. The nuclei that stain may be marginal cells. Occasionally staining in other nuclei around the isthmus is observed in larvae, along with some nuclei in the tail. The 5' upstream region of this construct is 2kb. Expressed in nuclei and sometimes accompanied by cytoplasmic staining.
Expr1431 In six comma-stage embryos, an average of 80.5 ( 2.5 s.d.) PHA-4-staining pharyngeal nuclei were counted by confocal microscopy (maximum count = 85). Analysis of later stage animals shows clearly that all six cells of the pharyngeal-intestinal valve contain PHA-4 protein. Thus the total number of potentially staining nuclei (pharynx + valve) should be equal to 86. all cells of the pharyngeal primordium (+ valve) contain the nuclear factor PHA-4. The same embryos also show nuclear PHA-4 in 6-8 rectal cells, including the two rectal valve cells and the three rectal epithelial cells. At the lima bean and comma stages of embryogenesis, a low but significant level of PHA-4 can be detected in the gut. pha-4 gene is expressed in pharyngeal precursor cells well before the formation of the pharynx primordium. PHA-4 can be detected immunologically in most but not all nuclei of the larval and adult pharynx. PHA-4 is detected in nuclei of all epithelial cells, muscle cells, marginal cells, gland cells and pharyngeal intestinal valve cells, but is detected only in about 8 of 19 neuronal nuclei. PHA-4 is also detected in one (comma stage) to several (pretzel stage and later) head cells outside of the pharynx. PHA-4 can also be detected in nuclei of the developing somatic gonad, including the distal tip cell and ventral uterine cells. nuclear
Expr11526 CYP33E2 promoter-driven expression of GFP occurred exclusively in the pharynx and, not visible in each individual, in the pharyngeal-intestinal valve of the nematodes. This type of strong pharynx-restricted expression was observed throughout larval development and in adult nematodes. Expression was most prominent in the pharyngeal pro- and meta-corpus. Confocal imaging suggested marginal, muscle, and/or epithelial cells as the major expression sites of the pCYP33E2::GFP construct within the pharynx. Radially, only marginal cell types are continuously organized with three-fold symmetry around the pharyngeal lumen. Imaginary cross sections derived from confocal imaging series of the pro- and meta-corpus indicated that the GFP reporter was expressed in the three marginal mc1 cells, but not in the pm3 and pm4 muscle cells. A further labelling of the mc2 and mc3 marginal cells in the isthmus and terminal bulb becomes then visible. Expression was observed in finger-like fluorescent structures that represent the interlocking extensions that hold marginal cells to muscles. Furthermore, an expression of pCYP33E2::GFP also in the epithelial e1, e2, and e3 cells seems most likely. Fluorescence might also correspond to pm2 muscle cells.

Feature : "myo-2.B207"   Expr11410 Multimers of a single oligonucleotide from the B fragment (B207) activate pharyngeal expression in a pattern very similar to that observed with the duplicated B fragment, with expression predominately in pharyngeal muscles m3, m4, m5 and m7. Unlike the larger B fragment, the B207 oligonucleotide occasionally activates additional expression in cells other than pharyngeal muscle (e.g. pharyngeal marginal cells, body wall musculature and intestine).
Feature : "myo-2.C"   Expr11414 The C+C enhancer (pOK17.21) activates frequent expression in all pharyngeal muscles. the C+C enhancer also activates expression in non-muscle cells in the pharynx. Expression induced by the C+C enhancer was observed in all pharyngeal cell types including gland cells, neurons and marginal cells as well as muscles. These results define C as an organ-specific subelement.
Most of the tissues expressing rcn-1 overlap with those observed in calcineurin GFP and by immunostaining, including lateral hypodermal cells, vulva muscle tissue, nerve cords, and diverse neuronal expression. Reporter gene fusion type not specified.   Expr2548 GFP analysis of the transformed animals revealed that rcn-1 is expressed from late embryonic stages to adulthood in diverse tissues, including lateral hypodermal cells, marginal cells of the pharynx, vulva epithelial cells, ventral and dorsal nerve cords and commissures, and various neurons in the anterior and posterior portions of the worm. Male C. elegans displayed rcn-1 expression in male tail structures including the diagonal muscles, sensory rays, and spicules. GFP expression analysis of a shorter 1.6 kb 50 upstream fragment of rcn-1 revealed vulva muscle expression in addition to the aforementioned expression patterns.
Feature : [ceh-22de199::pPD95.27]   Expr11805 The distal enhancer de199 can activate transcription in the pharyngeal muscles, although it exhibits distinct patterns of activity in other cell types. 5Xde199 activated reporter gene expression more broadly than the distal enhancer DE3 in most pharyngeal muscles and in marginal cells, with occasional expression in pharyngeal epithelial cells and in body wall muscles.
Feature : [ceh-22de209::pPD95.27]   Expr11806 The distal enhancer de209 can activate transcription in the pharyngeal muscles, although it exhibits distinct patterns of activity in other cell types. 5Xde209 activated reporter expression in a pattern similar to the distal enhancer DE3 in the pharyngeal muscles, with occasional expression in the m6 muscles, marginal cells, and pharyngeal-intestinal valve cells. The parental Dpes-10DlacZ reporter in pPD95.27 was occasionally expressed in posterior gut cells, with < 0.25 h-galactosidase expressing pharyngeal cells transformant.
Expr12323 sma-1:: GFP was expressed in all epithelial cells, as expected (hypodermis, excretory canal cell, intestine, pharyngeal muscles, and pharyngeal marginal cells) (McKeown et al. 1998) but was not detected in gland cells. Likewise, use of a nuclear-localized sma-1::GFP::HIS2B reporter did not indicate any expression in gland nuclei.
Feature: 'WBsf919537::pPD95.21'   Expr11811 The distal enhancer activated reporter gene expression both inside and outside the pharynx. In larvae and adults, expression was observed in the m3, m4, m5, and m7 pharyngeal muscles as well as pharyngeal marginal cells, epithelial cells, and neurons. Expression was also observed outside the pharynx in the body wall muscles and the ventral nerve cord. Distal enhancer activity initiated in the pharynx at the bean stage of embryogenesis near the time that the endogenous ceh-22 gene is first expressed.
Expr11294 Expressed in hypodermis, pharyngeal muscle and marginal cells, gut, some unidentified cells in the head (probably not neurons).
Expr11295 Fairly weak expression in most animals, strongest expression in young larvae, pharynx: marginal cells and pm6, gut, rectal gland cells, about 10-15 pairs of head neurons incl. some IL's, some motoneurons (DA/DB?), 2-3 pairs of tail neurons
Expr11297 Very weak expression, pharyngeal muscle and marginal cells, pharyngo-intestinal valve, 8-20 head neurons, 2 neurons in the tail, 2 unidentified tail cells (rectal gland?)
Expr11299 Expressed in body wall muscle, pharyngeal muscle and marginal cells, gut, distal tip cell, uterus, CAN, excretory cell (not visible on most images), coelomocytes, several neuronal and non-neuronal cells in head and tail, strong expression at all stages including embryos

Transgenes containing the VAB-19 cDNA under the control of the ajm-1 promoter, which is expressed in the epidermis and other epithelial tissues but not in muscles, also fully rescued vab-19 phenotypes, consistent with VAB-19 functioning in epidermal cells.   Expr2823 VAB-19::GFP was first detected in embryos at the onset of elongation. At this stage, VAB-19::GFP localization was diffuse in dorsal and ventral, but not in lateral, epidermal cells. During early elongation, VAB-19::GFP started to accumulate in muscle-adjacent regions of dorsal and ventral epidermal cells. During intermediate stages of elongation, VAB-19::GFP became localized to the parts of the epidermis adjacent to body wall muscles. Finally, during the late elongation, larval and adult stages, VAB-19::GFP localized to circumferential bands in epidermis adjacent to muscles and to mechanosensory neuron processes. Within these regions, the pattern of circumferential bands is interrupted by gaps where other neuronal processes intervene between muscles and the epidermis. This pattern of localization of VAB-19::GFP resembles that of epidermal attachment structures. VAB-19::GFP was also expressed in pharyngeal marginal cells. In contrast to Myotactin, which is localized to the outer basal surface of marginal cells, VAB-19::GFP was localized throughout the apical-basal axis, although it was concentrated at the apical and basal surfaces. Attachment structures contain several intermediate filament (IF) proteins, some of which are recognized by monoclonal antibody MH4. During early elongation, MH4 staining accumulated in a single patch in dorsal epidermal cells, whereas VAB-19::GFP remained diffuse. During the intermediate elongation stage, VAB-19::GFP and MH4 staining displayed more extensive colocalization. During later elongation and afterwards, VAB-19::GFP and MH4 staining fully co-localized to circumferential bands. The transmembrane protein Myotactin maintains localization of attachment structures to muscle-adjacent parts of the epidermis. Myotactin itself initially localizes to muscle-adjacent regions of the epidermis and later becomes organized into attachment structures. VAB-19::GFP colocalized with Myotactin before the twofold stage. Both Myotactin and VAB-19::GFP localized to circumferential bands. These bands are interrupted by gaps where neuronal processes intervene between muscle and epidermis. In contrast to Myotactin or VAB-19::GFP, MH4 staining is present or enhanced at such positions. These differences suggest that VAB-19 does not colocalize with all epidermal IFs but only those associated with muscle adjacent attachment structures.
Expr11304 Weak expression, gut, spermatheca, 2-6 head neurons. pharynx: muscle and marginal cells, pharyngo-intestinal valve, gut: weak expression in mainly anterior-most and posterior-most cells, some arcade cells, rectal gland cells, occasionally very weak expression in hyp (adults), 2 pairs of interneurons axons projecting into ventral nerve cord, 1 pair very weak, no longer visible in older animals
Expr12085 plr-1::GFP expression became visible in late gastrulation stage embryos. Strongest expression was detected in many cells in the tail region and by comma stage the most prominent expression was seen in body wall muscle cells. In early larval stages expression was detectable in a number of different tissues including the major hypodermal cells, muscle and marginal cells of the pharynx, the intestine (strongest in the anteriormost and posteriormost cells) as well as the anal depressor and stomatointestinal muscle. In the nervous system GFP expression was visible in a few neurons in head ganglia, many ventral cord motor neurons and several neurons in the tail ganglia including the PDA neuron. Based on the lack of commissures the motor neurons are likely of the VA, VB, VC and/or AS class. Based on the number of cells the majority (or even all) of these classes express GFP. In general expression was strongest in the tail region throughout development. This also held true for the motor neurons in the ventral cord, where GFP was strongest in the posterior-most cells and barely detectable in anterior motor neurons. Expression is maintained throughout larval development. In later larval stages expression is also seen occasionally in the distal tip cell of the developing gonad, the vulva and uterine muscle cells and the VC4 and VC5 neurons flanking the vulva. Expression in AVG, HSN or CAN neurons was not detectable with this reporter construct, but was detected in a previous study in HSN and CAN using a longer genomic construct (Moffat et al., 2014, Expr11480).
