Using transcriptional reporters, we found that the nrf-5 promoter (Pnrf-5) drove expression in body wall muscle cells. We also generated NRF-5 translational fusions with or without the secretion signal (Pnrf-5ssGFP::mNRF-5 and Pnrf-5GFP::mNRF-5). Secreted GFP::mNRF-5 (ssGFP::mNRF-5) displayed a diffuse staining pattern and associated with the basal surface of pharyngeal cells and the basal-lateral membranes of the intestine, whereas nonsecretory GFP::mNRF-5 labeled body wall muscle cells but not the intestine. These results suggest that NRF-5 is synthesized in and secreted from muscle cells to recognize apoptotic cells and associate with the basal surface of pharyngeal and intestine cells. |

The CED-3::GFP translational reporter was expressed mostly in embryos and the adult germline. In embryos, we often found an enrichment of GFP signal within the apoptotic corpses compared with the surrounding cells. In the germline, CED-3::GFP was expressed primarily in the oocyte region, with very weak to no expression in the mitotic and early meiotic zones. |
In oocytes, CED-3::GFP showed a significant enrichment in the nuclei, whereas, in the rest of the germline, we observed an accumulation of GFP signal in the perinuclear region. |

GFP::CED-11 was first detected in pre-bean-stage embryos, in which it was expressed broadly and had a diffuse cytoplasmic localization. By the twofold embryonic stage, GFP::CED-11 had a more restricted pattern inside the cell and also could be seen on the plasma membrane. GFP::CED- 11 was expressed in a few cells in the heads and tails of L1 larvae, where it was localized primarily to the plasma membrane. We detected GFP::CED-11 in puncta at the perimeter of apoptotic cells. |

In support of a role of ARX-2 in Q cell corpse clearance, ARX-2::GFP was recruited onto Q cell corpses at 37 ± 9 min (mean ± SD; n = 20) after Q cell birth, and the ARX-2::GFP rings lasted for 19 ± 9 min (n = 20). The corpse was completely degraded 34 ± 18 min (n = 20) after the disappearance of ARX-2::GFP, which suggests that ARX-2 may function during the engulfment of Q cell corpses but may not be involved in the final degradation inside the phagosome. |

A triple fluorescence animal that expresses ARX-2::TagRFP knock-in, GFP::WSP-1 knock-in, and the Q cell marker Plin-32::TagBFP shows that the majority of ARX-2:: TagRFP was recruited on the Q cell corpse at approximately the same time as GFP::WSP-1 and that the ARX-2::TagRFP fluorescence was largely co-localized with GFP::WSP-1 (n = 10). |
