Expression Pattern :

Primary Identifier  Expr11247 Subcellular Localization  From WBPaper00044072: Nuclear localization of DSB-1and DSB-2 is detected in the same region of the gonad, and their staining patterns on chromatin have a similar appearance. However, the relative intensity patterns of the two proteins differ during meiotic progression. Within the gonad,DSB-1 signal is detected on nuclei slightly before DSB-2 and has a stronger intensity early on, which then declines as nuclei progress through pachytene (except for the outlier nuclei); DSB-2 signal is weaker early on and peaks in intensity later than DSB-1 before eventually declining. Both proteins disappear from nuclei at the same time, and both localize to the same outlier nuclei. Within each nucleus, the intensity patterns on chromatin are also different, such that the DSB-1 and DSB-2 signals partially overlap but do not match each other. Immunofluorescence staining revealed that DSB-1 is absent from somatic nuclei, and specifically localizes to chromosomes during early meiotic prophase, while dsb-1 mutants showed only background staining. Accumulation of DSB-1 on chromosomes was first observed in nuclei marked by crescent-shaped DAPI-staining morphology, corresponding to the 'transition zone' (leptotene/zygotene), and disappeared at mid-pachytene. Chromosomal localization of DSB-1 preceded the appearance of RAD-51 foci, consistent with an early role for DSB-1 in meiotic recombination. Thus, the localization of DSB-1 to chromosomes corresponds to the period during which double strand breaks are likely to be generated. Most DSB-1 staining was associated with the periphery of DAPI-stained chromosomes rather than axes, suggesting that DSB-1 is primarily associated with chromatin loops.
